Birth Defect Amelioration and Placental Cytokine Expression in Mnu-exposed Dams Treated with Ifn-γ

چکیده
BACKGROUND: Methylnitrosourea (MNU), an alkylating agent derived from creatinine metabolism, is cytotoxic, genotoxic, and mutagenic. Mid-gestational exposure to MNU leads to distal limb defects in mice. Previous studies have shown that nonspecific maternal immune stimulation protects against MNU-induced teratogenesis. A role for immune-mediated placental improvement in this effect remains uncertain. METHODS: The immune system of timed-pregnant C57BL/6N and CD-1 mice was stimulated by gestation day (GD) 7 intraperitoneal (IP) injection with the cytokine, interferon-gamma (IFN-γ). A teratogenic dose of MNU was then administered by IP injection on the morning of GD 9 to disrupt distal limb formation. Fetal limb length, body length, digital deformities and placental integrity were evaluated on GD 14. RESULTS: The incidence of syndactyly, polydactyly, and interdigital webbing in MNU-exposed mice was decreased by maternal IFN-γ treatment. In C57BL/6N mice, these defects were reduced by 47%, 100%, and 63%, respectively; as compared to previous reports in CD-1 mice, by 39%, 71%, and 20% respectively. Administration of IFN-γ significantly diminished MNU-induced endothelial and trophoblast placental damage in both strains of

Cytokine Status in Ukrainian Children with Irritable Bowel Syndrome Residing in a Radioactive Contaminated Area
Background: The effect of low dose radiation on immune system is shown. Ionizing radiation can affect cytokine production and polarization of T helper cells. Objective: The current study focused on ionizing radiation in Ukrainian children residing in a contaminated area with clinical irritable bowel syndrome. Method: Our study included 75 rural children population aged 4-18 yrs, who lived in a ...

17 Inflammation during placental malaria (PM) is associated with low birth weight (LBW), 18 especially during first pregnancy, but the relative contribution of maternal or fetal factors 19 that mediate this effect remains unclear, and the role of IFN-γ has been controversial. We 20 examined the relationship of maternal and cord plasma levels of IFN-γ, TNF-α, IL-10, 21 ferritin and leptin to bir...
متن کاملExpression Pattern of Interferon-γ in Human Leukemic T Cell Lines Following Treatment with Phytoheamagglutinin, phorbol myristate acetate and Lipopolysaccharide
Background: As a T helper type 1 (Th1) derived cytokine, Interferon gamma (IFN-γ) is an important regulator of inflammatory immune responses. Furthermore, IFN-γ plays an essential role in defense against tumors and intracellular pathogens. This study was designed to assess the pattern of IFN-γ production in human leukemic (Jurkat and Molt-4) T cell lines in vitro. Methods: Ju...

متن کاملImmunopotentiotor Effect of α-Tocopherol on Cytokine Expression in the Lymphocytes in the Elderly People
Background: Aging is associated with attenuation of immune responses. Studies show that old people are vulnerable to infectious diseases such as influenza. α-Tocopherol as an immunomodulator affects immune responses. In the present study, the effect of α-tocopherol, on lymphocyte responses i.e. interferon-gamma (IFN-γ), Tumor Necrosis Factor-alpha (TNF-α), and nuclear factor-κB (NF-κB) in elde...
